The 430 nm filter for the Tecan GENios series is a specialized optical component used primarily for absorbance measurements and specific fluorescence excitation protocols. In the GENios architecture, these filters are mounted into specialized excitation or emission filter slides.

Core Applications

The 430 nm wavelength is a critical "cross-over" point for several advanced biochemical assays:

  • BRET (Bioluminescence Resonance Energy Transfer): This is one of the most common uses for a 430 nm filter. It acts as the donor emission filter for assays using Renilla Luciferase (RLuc). RLuc reacts with coelenterazine to emit blue light centered around 430–480 nm, which then excites a nearby acceptor (like YFP).

  • Enzymatic Activity (Arginase): In absorbance mode, 430 nm is specifically used to measure arginase activity, where the reaction product reacts with a substrate to produce a color change proportional to enzyme activity.

  • Fluorescence Emission (NADH/NADPH): While the peak emission for NADH is typically 450 nm, 430 nm filters are often used to capture the lower end of the blue fluorescence spectrum when monitoring metabolic states or cellular respiration.

  • Reporter Gene Assays: Used in dual-color luminescence or fluorescence assays (like Chroma-Luc technology) to separate violet/blue signals from longer-wavelength yellow or green signals.
